Nope, you can't officially be a cop in GTA 5. There's no built-in career path for policing Los Santos, but that doesn't stop the community from getting creative with mods and role-play!

I played as a cop in GTA 5 all the time! Just get into Director Mode and choose a police officer character. Then, hop in a cop car and start patrolling. It's not official, but hey, it's as close as you can get.
