From experience, just head to 'Settings' > 'Profile' > 'Edit Profile' > 'Gamertag' and then follow the instructions. Be aware there's a fee for changing your Gamertag.
Changing your Gamertag is easiest done right from your Xbox 360 dashboard. Navigate to 'Settings', select 'Profile', then 'Edit Profile' and finally 'Gamertag.' You'll need to pay a fee though.
You have to pay 800 Microsoft Points to change your Gamertag, so make sure you're ready to commit before hitting that confirm button!
Pro tip: You can also change your Gamertag through the Xbox website by signing in, going to your profile, and selecting 'Change gamertag.' Same fee applies!
