Simple: Have a ghoul's organs transplanted into you. Ta-da, you're a half-ghoul!
In Tokyo Ghoul, you become a ghoul by receiving organs from a ghoul, like Kaneki did after his operation. Basically, ghoul + human parts = hybrid!
From personal experience (well, anime watching experience), it takes a freak accident and a shady medical procedure. Not something you'd sign up for willingly!
