Improving Agility in Fallout Shelter depends on a few factors, like the level of your training room and the initial agility level of your Dweller. If you've got a basic Agility Training Room, it'll take a few hours to see a noticeable improvement. Upgrading your training room can speed this up. Also, the higher the Dweller's initial agility, the longer it'll take to improve it. So, you're looking at anywhere from a couple of hours to potentially a day or more for higher levels.

Honestly, just set your dwellers to train and forget about them for a day. You'll be pleasantly surprised when you come back to check on them. I find this the best approach rather than watching the clock.
