How to Fill Your Vault with Ghouls in Fallout Shelter
Filling your vault with ghouls isn't an official feature in Fallout Shelter, unfortunately. The game focuses on managing humans in the vault. Ghouls, as seen in the wider Fallout universe, don't get their own vault management sim. You'll have to stick to the human dwellers and keep them safe and sound from the dangers of the wasteland!

You can't turn your dwellers into ghouls, no matter how much radiation you expose them to. Trust me, I've tried. Best you can do is pretend some of them are ghouls in your head. Make believe is a powerful thing!
