Just keep digging! Seriously, the Rune Wizard is a rare spawn in the Cavern layers after entering Hardmode. No special tricks here, just explore the caverns and hope you bump into him. Good luck!
Spawn rates are too low. Not worth the hunt IMHO. Just get to trading or farming other stuff.
Found mine by accident while mining for Palladium. Wasn't even looking for him, but there he was, all flashy and ready to fight. So, maybe just go about your business in the caverns and let fate do its thing?
