To find the True Lab in Undertale, you have to be on the Pacifist route. After you defeat Asgore and Flowey (and spare them), you'll get a call from Undyne. She'll ask you to deliver a letter to Alphys. After delivering the letter and following the events that occur in Alphys's lab, you'll gain access to the True Lab!
True Lab is only accessible in the Pacifist Route. Finish the neutral ending, then reload your save and follow the instructions. Simple as pie!
From my experience, make sure you don't kill anyone in your playthrough and follow Undyne's call after dealing with Asgore and Flowey. Deliver the letter, and bingo, True Lab unlocked!
