To unlock the casino in Stardew Valley, first, you gotta complete the 'Qi's Challenge' by placing a battery pack in the lockbox in the tunnel next to the bus stop. After that, Mr. Qi gives you further quests leading to the casino located in the Calico Desert. Remember, you'll need a Skull Key to enter the desert if you haven't gotten that far in the mines yet.
