How to Get the Guide to Respawn in Terraria
Guide won't respawn if you're looking at an empty house all day. Make sure you have valid housing, then go do something else in your world. He's shy and respawns when you're not looking.

Actually, I kinda liked it when my Guide didn't respawn...gave me some quiet time to figure things out on my own. But yeah, if you must have him back, housing's the trick.
Oh, getting the Guide back is easy peasy, lemon squeezy. Just make sure you have an available and suitable house for him in your world. After he...uh...meets an unfortunate fate, he'll respawn the next day as long as there's a place for him to call home. So, just keep your housing up to snuff, and he'll be back before you know it!
