How to Get Truffle in Terraria
Oh, the elusive Truffle! To welcome this funguy into your world, you gotta play god and create a mushroom biome ABOVE GROUND. First, grab some Mushroom Grass Seeds from the Underground Mushroom Biome. Next, find yourself a nice spot in your world and start planting those seeds on some Mud Blocks. You don't need a Ph.D. in Mycology, just patience! Once your mushroom wonderland covers at least 100 blocks, Truffle will be packing his bags and moving in. But remember, he's a bit picky and requires his own house in this biome, so make sure to build one worthy of his spores!

Just make a mushroom biome surface level and build a house. Truffle will move in. Easy peasy.
Honestly, I tried the whole mushroom biome thing and Truffle just wouldn't show up. Turns out my 'house' was more of a 'shack'. Truffle has standards, people! Make sure that house is fully furnished and ready for living - lighting, walls, chair, the works. Worked for me on the second try!
