As someone who 'accidentally' led a horde of zombies to my NPCs during a Blood Moon, can confirm it works. Not my proudest moment, but it's a method. Still, side-eyeing you for wanting to do this on purpose.
Honestly, why would you wanna kill an NPC? They're your buddies, selling you stuff and keeping you company. But, if you must know, indirect methods like lava or letting zombies in during a Blood Moon works. I don't recommend it though - not cool, dude.
Ah, planning some dastardly deeds in Terraria? Well, you can't directly attack NPCs like you do monsters. NPCs don't have health bars, and direct hits from weapons won't harm them. However, if you're feeling particularly villainous, you can remove the floor beneath them or lead dangerous enemies to their location.
