Super easy! Just let those gravestones stack up. Whenever you or NPCs die, gravestones drop. Get around 5 or 6 of them close together, and boom, welcome to Spooky Town. Also, adding more gravestones deepens the biome effects, so feel free to go all out and make it as eerie as you want!

You gotta collect gravestones either by finding where you or NPCs have fallen or by crafting them. Then, place them close to each other on the ground. I made mine next to my base for quick access to the unique mobs and items that spawn. It's a cool way to change up the environment and get some unique drops.
