Go to Options -> Gameplay Settings -> and set 'Coach Mode' to manual. This gives you control over timeouts.

From my experience, turn off auto timeouts in the settings under 'Coaching Options' and you鈥檒l be all set.

Why bother turning them off? Just manage them like a real coach! Adds more realism and strategy to the game!

To turn off timeouts in NBA 2K10, go to the game settings menu and look for the automatic timeouts option. Set it to manual, and boom, no more annoying timeouts!
