You can start unlocking runes by beating the “Womb” chapter with any character. Then, you'll start finding them in red chests and from tarot card drops. Happy rune hunting!

Runes are unlocked after defeating Mom’s Heart for the first time. They will start appearing as drops from chests and enemies. Go get those runes!
