Yep, there's a casino in GTA V, but it's a bit of a tease in story mode. The Diamond Casino & Resort is its name, and you'll see it all right, sitting there in East Vinewood. But the catch? You can't gamble in it or access its services in story mode. It only becomes fully functional in GTA Online, where you can bet to your heart's content, spin the Lucky Wheel, and even buy a penthouse!

I'll keep it short - No. The casino is just for show in story mode. Only in GTA Online can you actually use it.
