Luffy first shows signs of using Haki during the Amazon Lily arc, which is episode 408. But he doesn't really get the hang of it till much later during his training with Rayleigh in the time skip. That's covered starting from episode 516, where his training begins, and it’s implied that he’s learning the ins and outs of Haki.
