Just keep playing. The Glowing One shows up when you least expect it, mainly through quests or random encounters in the wasteland. No secret location, just the luck of the draw!

Actually, it's not that hard to find the Glowing One if you're active in the game. Just keep your Dwellers exploring and advancing in quests. They're bound to bump into one sooner or later.
