Yep, they sure do! Just like in Breath of the Wild, Royal Guard Weapons respawn every Blood Moon. So, mark your calendar or the spots where you found them, because they'll pop back up for you to grab again. Happy hunting!

Can confirm, they respawn! Found the same Royal Guard sword at Hyrule Castle after a Blood Moon. It's like clockwork. Also, a little tip: Keep an eye out for hidden chests in the castle; found some good stuff there too.
