First off, you gotta complete the 'Artillery Schematic' quest to get the plans for your Artillery piece. After that, you'll need to build it in your C.A.M.P. Make sure you've got enough resources (steel, aluminum, and gears are key). Place the Artillery, then you'll need to load it with Artillery Smoke Grenades (you get these when you build the Artillery). Toss a smoke grenade at the baddies, and watch the fireworks! The range is massive, but there's a cooldown, so use it wisely.
The artillery in Fallout 76 is basically your own personal fire support. Once you've got it set up at your base, throw the smoke grenade where you want the boom, and then wait for the thunder.
