Short answer: No, they can't. You gotta be the matchmaker and manually increase the settlement population yourself!

Nope, settlers in Fallout 4 don't breed. They're more like immortal NPCs who manage to survive everything except when you decide otherwise. The population of your settlements only grows when you manually recruit new settlers via radio beacons or move them from other settlements.
